题目: Fluorescent and bioluminescent proteins in live cell and deep-tissue imaging

Jun Chu received his Ph.D. degree in Biomedical Engineering from Wuhan National Laboratory for Optoelectronics at Huazhong University of Science and Technology in 2009, did his postdoctoral researchin Jeanny Hardy lab at University of Massachusetts, Amherstfrom 2009 to 2010 and Michael Lin lab at Stanford University from 2010 to 2015. He has developedseveral novel fluorescent proteins (FPs) and FP-based biosensors. 1) afar-red bimolecular fluorescence complementation system (BiFC) that works very well under cell physiological conditions, which could be used for monitoring protein-protein interaction in vivo; 2)a GFP-based caspase-3 indicator(CA-GFP) with large dynamic range, which potentially can be used for visualization of cell’s spontaneous apoptosis during embryo and neuron development; 3) a brightest far-red FP, mCardinal, providing a useful tool for optical imaging in vivo; 4)a large Stokes shift orange FP (CyOFP), which can be excited together with GFP using single wavelength, allowing simultaneous visualization of two molecular events under two photon excitation in vivo; and 5) a green FP-red FP based FRET (fluorescence resonance energy transfer) pair (Clover3-mRuby3) with large dynamic range, which is useful for detecting weak or transient biochemical responses in living cells. He has first-authored and co-authored more than 15 journal and conference articles including one Nature Methods, 2 U.S. patents and one invited book chapter. He is a member of Biophysical Society and Protein Society.  His current research interests include engineering of novel infra-red FPs for optical imaging and red-absorbing chromoproteins for photoacoustic imaging, engineering of novel large Stokes shift FPs for two photon imaging, improving existing FRET pairs and development of FRET biosensors for kinases, and development of new optogentic tools for perturbing the signaling pathway in living cells.

Fluorescent proteins (FPs) and bioluminescent proteins (BLPs) have been widely used as molecular and cellular reporters in living organisms. However, existing FPs and BLPs suffer low brightness and sensitivity, limiting their applications in detection of biochemical processes with subtle change in cells or monitoring biological events in small animals. Here we report several new FPs and BLPs obtained by structure-guided mutagenesis and their applications in living cells and animals.
