  Title: Kilohertz two-photon fluorescence microscopy imaging of neural activity in vivo

  Abstract:

  Calcium imaging using two-photon fluorescence microscopy has been routinely used to dissect the complex mechanisms of neural activity in small behaving animals. The calcium signal, however, is too slow to follow fast spike trains and cannot report subthreshold activities. Voltage imaging monitors the electrical signal and holds the key to address these limitations. Using an all-optical laser scanning technique, we developed a two-photon fluorescence microscope with speed up to 1000 frames per second, a frame rate suitable for voltage imaging. With existing genetically encoded voltage indicators, this imaging technique enabled us to visualize spikes and subthreshold activities of neuronal populations in head-fixed awake mice.
